If it starts on second try, I doubt it is the car battery. But you can check the car battery after it rests with a volt meter. If the voltage is less than 12 volts, you may have a bad cell in the battery.
Do you have key start or push button start? If push button start, the battery in the key fob may be going bad (Toyota key fob batteries seem to go bad faster than others).
Also check the car battery terminals for corrosion.
Finally, I wonder is something is going on with the engine immobilizer system. The engine immobilizer system does not allow the car to start if an authorized key is not present. The car reads a chip within the key to see if that key is authorized. The chip in the key does not use the key battery; it works somewhat like the inventory control tags at stores that set off an alarm when you try to exit a store without paying for the item.
