I believe the difference is in the valve stem. You can get sensors that have the valve stem as part of the sensor, and then there are sensors that plug into a separate valve stem that looks like the old fashioned rubber valve stems. So the answer is both will fit, just pick the one that has the type of valve stem you want.
Not quite sure on the 4Runner, but for my RAV4, as long as replacement wheels have OEM TPMS, there is a setting in the vehicle settings for "change tires". So when you change out your summer tires for winter tires, you select that on the settings, then the car will automatically sense the new TPMS after you drive a few miles. (The tires have to rotate for some time for the TPMS to wake up).
