I have for the 4.0 4th gen. I'd say the biggest difference is in throttle mapping through engine expected torque or similar maps, and transmission tuning. That said, I'm still not fully happy with the transmission tune and may just make my own tune instead. As a professional tuner they have access to the full range of maps (vftuner won't give access to the layman), but I don't know that their tweakings in the engine management actually made much difference. You will have access to throttle parameters and shift points, so I would recommend trying your own hand here first. You can look on the tacoma boards for guidance.
