if you are referring to installing a supercharger on to a 3rd gen then yes it makes taking out the spark plugs on the drivers side cylinder bank a PITA. also you need to take into account what a supercharger will do to your engine. you are basically shoving more air into the engine than what it was designed for (granted the 5VZ is basically bulletproof and can take the beating), so you have to account for the fact that the factory fuel map isnt able to adjust properly for the amount of air being jammed into the cylinders, which can lead to knocking and a lean air fuel mixture, both of which can seriously damage your engine. a common way to add more fuel is a seventh injector system. also i don’t know your budget but these superchargers can go for well over $2500, plus there are other parts that you need, some of which may be hard to find. i am not trying to discourage you from installing one because you totally should (they are sick as f***) but i am just trying to prepare you for a couple of obstacles you could run into. check this thread out:
