Yes, 4th Gen V6 4Runners are able to be in 2WD mode as Front has vacuum disconnect. I'd advocate for 4WD version, makes life much easier in the winter, especially out of the city. If you travel gravel roads or using it to tow a boat, 4WD is beneficial as well.
