will the rotor point to number one cyl everytime the crankshaft mark is on tdc or do i need to remove the plug to see when it is tdc?

I sprayed starting fluid into the throttle body and still no start. Would this eliminate the question of injectors firing?

how would I be able to tell? I smell gas after 20 sec of cranking, but the one plug I pulled was dry.
