You don’t have to use anti seize on the plugs. The MFR doesn’t recommend them because the threads/metal are coated with anti seize. If you do, they will seize up when you remove them. Toyota doesn’t recommend the iridium plugs for the V6 and change them out every 30k miles.
You're right, coils USUALLY work or dont work. But overtime they loose their efficiency. Coils produce thousands of volts and a lot of heat. With 250k miles on the original coils and the heat from engine will degrade the coils. Ive owned my 4Runner almost 16 year's now and I change my plugs...

I had the same issue and ended up replacing the drivers side CAT PO430. I was told their was only one California Approved CAT available at the time for the 4Runner. New CAT and labor was $1000.
You should check the gaskets after the CAT to see if they are bad just to confirm its not the gasket.

Its a common issue with the 4th Gen and 5th Gen. Possible issue could be its coming from the rubber hangers for the exhaust/muffler. I have over 253k miles on my 03 and I still get the burning rubber smell now and then.
