Driving cross country, need MPG's!

The only thing that will be cost effective on your list is to air the tires up. I doubt you'll realize a return on your investment for any of the other things. If you need new plugs, then by all means get them, but don't switch them out just in the hopes of getting better fuel economy during your trip. You'll never recover the cost in a 3,000 mile trip!

I have my tires aired up to 42 in the rear and 44 in the front, right now should i let some air out?

It has been my experience that my 4Runner's MPG sweet spot is around 55-60mph.
Unfortunately, that speed is too slow for many driving situations.
I keep my tires close to the maximum pressure specified by the tire manufacturer and Toyota.
Cruise control (when not in heavy traffic) is a must.
I only keep essentials in my 4Runner to avoid hauling excess weight around.
Avoid unnecessary stops and idling when possible. Always runs with the windows up at highway speeds and use the A/C if necessary.
Avoid using a rooftop carrier whenever possible.

My 4Runner is a V8 4WD. I can easily get 20-21 MPG on the highway. The closer I can drive to the sweetspot speed of 55-60 mph the easier it is to get that kind of MPG.
