that the a car like the Lexus RX330 or RX350 fuel cost is same or worse than the V6 4Runner.

Given that the 4runner is heavier, wider tires, truck based SUV, boxy shape, etc. It would be cost more on fuel than a Lexus RX. I know partly the reason is because the Lexus uses premium fuel but had no idea it cost more to operate as far as fuel goes.

My father in law has an rx350 and it has a turbo. It's a 2011 or 2012 though. Not sure when they started

Even if they did you are not comparing apples to apples, my comparison was of similar years but a 2012 is a lot newer. Besides a 2012 with or without turbo if they exist would get better gas mileage than a 2006 4runner
 
My previous car was a 2000 rx300 I averaged 15 mpg on a 16 gallon tank had a 250 mile range max. So the switch to the 4runner wasn't painful at all
