Not as interesting as the title sounds...

I have somewhat of a silly question, but I want to try to follow "best practice" when it comes to participating in the classified sections. If I am looking to sell an item, and need to give a "quote" on shipping the item to a specific zip code, what's the best way to go about this? Is it simply finding a kind employee at a ups/fedex/usps store or is there a better way?

Both USPS and UPS have online price calculators you can use.

USPS has flat rate boxes. The same price no matter where you send it.

Much easier, IMO, to just add $20 bucks or so to your price and advertise it as shipped. It may cost slightly more or less in actuality, but you won't have to calculate shipping for all the window-shopping bozos.
