Help 2006 will turn over but won't start

Got a 2006 sport v8 4R, 74500 miles on it...it cranks but won't start unless you mess with the key in the ignition and when it does start it runs fine. No warning lights come on, checked all the fuses. Also it is chiming when the door is open without a key in the ignition and it won't let me lock it with the remote. Even my roommate who is a mechanic is baffled.

Hmmmm... I cannot understand how it could "turn over" without ignition, and then fiddling with the ignition switch would suddenly cause the engine to fire up. Traditional thought would be no fuel or no spark but that would clearly not be the case here. Sounds like possibly a transponder issue within the key itself? I just cannot remember if the lack of a transponder causes no turn over or no ignition. Regardless, that is where I would start.

We've tried both keys and I even have an after factory remote start. Remote start even does it which should bypass the ignition switch if that was bad. Once we finally get it started, everything is fine, everything works normal and there are no warning lights that come on. No warning lights come on period and everything gets power.

Hmmm. Maybe the culprit is your aftermarket remote start. That would be my guess.

I'm guessing it is whatever the unit that receives the RF signal from the chip in your key.
It works fine with the remote start cause that has a unit which bypasses the search for the RF signal from the key.

Maybe someone else can shed more light on the unit that receives the signal?

I've had the remote start for over a year and never had any problem. The remote start will not start it, it gives us the same issue, turns over but never starts. Sprayed some fuel starter in and it started right up and runs normal, we can get 2 or 3 starts from it then the same issue happens. This is driving me crazy

Sorry, I misread it and thought you mean it starts properly with the remote start. Can you describe what you mean by "mess with the key"
The fuel starter starts it each time? if it was once, I'd say coincidense...

Also, for what it's worth, I had a few Honda/Acura's that did that when the weather got hot outside. It was a weak solder point in the 'main relay' Do our trucks have a main relay?

Wiggling the key around in the ignition as you're trying to get it to start seemed to work and also every time we sprayed the fuel starter into the throttle body it started right up and also a few times after. Nothing seems to be "wrong" with it bc once we do get it started it runs fine just like normal. I can't understand why it keeps chiming like the key is in the ignition with the door open when there is no key in it. I don't know if that issue is related or not.

If it starts right up and runs with the starting fluid then it sounds like a fuel delivery issue....fuel pressure test would be first on my list but honestly I am not sure where the service port is....anyone else?

No service port, both fuel lines (return and feed) are near the brake master cylinder. The are quick disconnect style fittings.

The fact that the vehicle acts like it has a key in the ignition when there isn't one makes me suspect there's a wiring fault with the aftermarket remote start, whether or not it hasn't caused problems in the past year doesn't mean it won't. I've seen aftermarket alarms installed "properly" by the dealership or shop develop issues later down the line rather than immediately.

But what you're saying is once the vehicle "catches" (in this case when you spray starter fluid) it runs fine, it just takes the starter fluid to get it going?

I'm going to agree with most of the others that it is the Remote start, however, it could be the starter. Maybe connections are loose, and jiggling the key moves the connection closer, allowing for current to flow.
