Just leave it in drive or go to 4th.

I'd leave it in drive. Even with "just" the V6, the Runner should barely even notice it's back there. Maybe drop it down to 4th in the mountains, but I'm betting drive should be fine.

You have a 5 speed? I would guess the V6 won't have any issues being in drive. If you start a steeper grade at high speed (highway speeds) and it downshifts, you'll know for the next hill. It would be a suprise if your transmission got hot at all. 1000lbs and under is a non-issue
