Question on non sealed transmisison

Guys, it is so strange I could not find anywhere in manual when to check transmission fluid on 2003 4runner. Should I check with engine idling or not? The dipstick has cold and hot marks, but nowhere it says should I check with engine turned off or running.
I would appreciate any information.

From the '03 FSM:
1. CHECK THE FLUID LEVEL
HINT:
Drive the vehicle so that the engine and transmission are at normal
operating temperature.
Fluid temperature: 70 - 80
 
All tranny's that I know of that you can check yourself (via dipstick) require that your vehicle be at full operating temperature and the vehicle in park after what Chris has explained. Remember, transmissions take a little while to warm up, so just don't go run around the block and soon as your engine is at temp assume the tranny is. They usually take much more. Do some stop and go driving around town for about 30 minutes or so to get it up then check it. Hope these inputs help!
 
Guys thans a lot! This means I am missing a quart. I serviced transmission at the dealer and they did not put enoght fluid. Now when running in park it hardly reaches cold mark.
