That's a fair amount of weight. Probably would upgrade to load range C or E from passenger. Which is what it seems you are already looking at. We haul a Jayco 165RB empty it's 3000. Loaded it's close to 5.

The stock tires are rated to tow everything the stock 4Runner is rated to tow. The 5th Gen (with V6) is rated for 5,000-lb; I believe earlier gens were rated for higher loads. If you put your vehicle specs in your sig we will automatically have more info.
When it comes to upgrades for heavy trailers, a 4,000lb (2-ton) gross weight loaded trailer is relatively light. Basically the equivalent of towing a full-sized passenger car on a front-wheel dolly. If you do a lot of trailer pulling, then maybe upgrade to a tougher C or E rated LT tires as mentioned above. Otherwise, go with whatever tire suits your daily needs.

I have used several tires for towing in a Ram 3500 with a Cummins. The Michelin LTX M/S offered the best year round traction, but the LTX A/S offered slightly better fuel economy and tread wear. Due to where I live, I chose to use the M/S.
Note that I ran probably 10 different tires in that truck including all terrain, mud terrain, and all season LT tires on it. Towing or not, your most important factor is choosing a tire of the appropriate load rating with the tread most appropriately suited to your typical driving environment. I found during 7 years of towing 230 miles per day that having the load properly balanced on the trailer makes a much greater difference in safely hauling the load.
I'm about to take a 7,400 mile road trip with a family of 6 and a cargo box on top. I'm getting new tires, likely BFG KO2's, largely because I plan on hitting up some dirt roads and trails while out West.
My advice to you is to make sure you get tires rated at least as high as the factory tires and are well suited to your daily drive.

LT245/75r16 Michelin LTX M/S Load range E is perfect for the trails here and has the plys to tow.
