Vibration is driving me crazy!

It could be your tranny mount, but honestly when you have one mount go bad, the rest follow shortly after as it puts more stress on them when one fails. If you have had an oil leak it can erode the material the mounts are made of and cause they to pretty much fall apart. If you are going to replace one, you might as well replace them all at once

I'm an expert in my own mind and a newb with the 4runner but I have had motor mounts fail every year on our ex-Mercury Villager and the hallmark sign of a failed motor mount was to hold your foot on the brakes and while in D or R and goose the throttle you would be able to see the engine rise and heave further than it should. A search of youtube should show you what to look for.
Have you thought about a failing U-joint. When you say the vibration hits at a specific MPH I'm thinking driveline imbalance not engine.

This just started for me today to, a squeek right under my feet. I just got new sliders welded on, so I initially thought it was that, but they are solid. I'm thinking its the extra weight of the sliders and causing the body mounts to sqeek, perhaps now the motor mounts. Driving me crazy.
Do you feel the vibration when the 4runner is at stop? If you get a vibration when driving it is either from unbalanced wheels or drive line.

Squeek? that doesnt sound like a mount. If its mostly from Idle to 1500rpm it could be a Flex Plate fracture.

Check your U-joints and make sure the slip yoke is greased.

Can you provide more info on the location of the flex plate area? Not familiar.
I think the sliders weight might have created tension on some joint or bearing, or a part of the sliders brakets is vibrating against other metal.
I might also try loosening the body mount bolt and retension.

I've got the same vibration, it will start while I'm going through a gear and stop when it finally shifts.

To the OP in the slight chance he looks back at this thread and hasnt solved his problem. If your fan shroud is still intact then chances are your motor mount isn't completely gone. I'm thinking driveshaft imbalance, ujoint gone bad or in need of grease. Id check the grease first.
I cant remember if our driveshafts are supposed to be in or out of phase off the top of my head. Could be worth checking in to anyone hasn't owned their truck long...no telling what the previous owner could have to done.
