What is this leak/build up?

Actually the valve cover leaking on higher mileage 2UZ-FEs is about the most common leak you'll see next to the factory water pump leaking by the first timing belt change interval (usually the second pump you put on there doesn't end up leaking again). The engines are really good about staying dry/not leaking excessively over time like other vehicles.

Valve Cover Gaskets are easy to do on the 2UZ-FE and the part is fairly inexpensive. Good weekend warrior skill building job. :)
 
They will probably be loose if you check the torque, but will keep leaking after you tighten them.

I tried switching to full synthetic on mine with 250,000 miles. After about 2 weeks I started to notice oil leaking from around the bell housing. After 2 months it was leaving puddles everywhere I parked it. I switched back to conventional oil and the leak has since dried up

Something to consider is that it's not so much the oil's fault as much as it's a symptom of using a better quality oil.

It sounds like the rear main may be leaking (and probably has been for a while), switching to the synthetic oil probably meant that much of the gunk that was "sealing" that leak up was being cleaned out; that combined with the more uniform size of synthetic oil molecules let them seep out more readily than conventional oil does.

Sealing technology has come a long way from the days of cork gaskets and simple rubberized foam in terms of oil interactions. So the urban myth of synthetic oils causing seals to leak is less because it breaks down the seal, but rather that it tends to clean out the engine a bit better and has a more uniform size as a microscopic level that lets it fit in smaller spaces and thus find leaks better.
