Locker Questions

I just recently bought a 2000 SR5 with a 5speed. it doesnt have a rear locker but i want to get one installed. I am thinking about getting a factory locker from toyota or the Eaton E-Locker. Any suggestions and would i need to change the gears and ring and pinions. Also i have heard some people talk about a center diff locker, what is the purpose of this? Thanks
