If you want something more comfortable just sell yours and get a 4th gen. Or a used Lexus RX

If you want it to be quieter and more comfortable, sound deaden everything and do new stock suspension, road tires, and poly body mount bushings

I don't know what year Avalon he has, but that checks off all the boxes you mentioned. MPG will be about 30 highway. Quiet and comfortable? Hell yeah, it's essentially a rebadged Lexus. It's a fantastic car. You said you don't take the 4Runner off road, so you don't need 4WD. The question is, how do you feel about owning an Avalon?
