How big of a difference will I notice in ride quality from an E rated tire to a P rated tire if the tire is stock size (265/70/16)? I spend most of my time on the highway, so I want the most comfortable ride possible....but I also want an aggressive looking tread because it looks cool.

I hate to hear that. The KO2s are my favorite looking AT tire.

Yeah, they look good, but I was very unimpressed with the ride quality. I'm also running 30psi in them.

I'm pretty sure you can get the KO2 in C load rating in that size.
I have the Falken Wildpeak AT3W's on my Runner in E Load, just got them today so I can't speak much for how they ride on a daily basis but initial opinion is they are pretty smooth for an E. They also make these ina P rated tire. They look fairly aggressive.

I have E rated KO2s and they don't bother me at all. I am sure there are softer tires out there, but I don't feel the overwhelming desire to ditch my KO2's any time soon. But if you like the look, do as others have suggested and call up discount tire and see if they offer the KO2's in your size in the C rating.

I am running KO's in a 31x10.5 15 C rated and it drives very nice in the highway. Road noise is minimal and it aborbs the bumps well. I am very happy with them.
