I have a 2006 V6 with switchable 2 wheel and 4 wheel drive modes. To keep things lubricated properly the manual suggest that the 4 wheel drive mode be used at least once a month for a few hours or so. Ok I get that part but what about all the other modes. For example is it necessary to drive in 4Lo mode every now and then. Is it neccessaryto lock the diff every now and then?

Yes absolutely necessary to drop it into 4L unless you want to replace a very pricey actuator. Just run a cycle from 4Hi to 4L then lock it up and then undo the process. Do not see it being necessary to run this for "a few hours" but necessary to cycle through the steps.

Engaging the 4hi is necessary to ensure the rotating components in the transfer case and the front diff get a chance to stay lubricated. Engaging the other modes like you mentioned dont impact the rotating components, they splash lubricate in just 4hi. Like others have mentioned, engaging these modes is still a good idea because the actuators are a use it or lose it piece if gear.

Way more fun to lock the diff on some sand or a muddy hill.
I found a big mud pit near me on Friday and it was irresistible. The rural-suburbs of Boston have NO worthy wheelin' sanctuaries. Gotta take what you can get! Got my truck all caked in dirt, it was beautiful haha.
