cylinder misfire

So I have a 2000 4runner 4wd drive with the v6 engine. Cylinders 1 and 6 are misfiring. Its only an issue at speeds above 60 MPH. I've been told it is probably the coil pack or the leads. If anyone has any insight on this issue any advice would be greatly appreciated thanks

Start by swapping the coils around and see if it misfires on new cylinders. New plug wires are always good.
