My TPMS light has been on forever even though they're all new, except the one in the flat spare that doesn't even match anymore. Will the light go off if I simply remove the spare from under the rig?

No, because the computer is getting no signal from the 5th sensor.
My TPMS light has been on since I switched to FJ cruiser rims and didnt transfer the valves due to one of them having a close to dead battery, causimg the indicator to flash.
If you look for the DIY fix, its cheap and quick.

Yes I did this. All you have to do is jump two wires to disable the system. I jumped a switch in also to make it switchable on/off.

I just purchased a 2008 4R that has had the TPMS bypass done. Is this the current thinking, to just do without TPMS? Did it not work reliably before?
I was thinking about getting new TPMS monitors and trying to see if I can get the system working again. But I'm having my doubts. Does anyone bother? Or is that considered a waste?

I've never seen the TPMS work in the 4R. Does it give you a pressure (psi) reading in each tire or is it just a general alert to low pressure somewhere? In other words a true gauge or an idiot light?
My 2008 4R TPMS has been working fine since day 1. After about 10 years the batteries started to die and had all 4 replaced. My new stereo (Kenwood DDX6906S) has a procedure to match the individual tires up to a display of the vehicle so I know which tire is low. Seems to trigger at about 25psi. Display shows actual PSI of each tire.

Yes, the TPMS light will go off if you remove and keep the spare off your rig.
It's possible that if you didn't change your wheels/TPMS sensors, that the battery is dying on the sensor. Either replace the TPMS sensor, wait until you need to get new tires to insert new TPMS sensor, put tape over the dummy light or just bypass it (Write-up: How to Disable (Bypass) the TPMS)
I have a '04 V6 SR5, and one of my tires had a slow leak due to the corrosion on one rim. The TPMS light would illuminate (solid) once a week, so it helped me in that sense. Once I had some bead sealer put on and got that problem fixed, one of the batteries started dying, and the light would blink consistently. At that point, I upgraded my rims and tires and opted for no sensors and have been living with a solid TPMS light. My car does not have any way of displaying each tire pressure, even with a OBD reader and the Torque App.

Thanks gents, I'll drop the flat/dead spare and toss it in a dumpster. As for the light, I'd prefer to keep it. When you run over a piece of metal on the highway and the light comes on it gives you fair warning to get off the road. Don't ask. (my TRD pros and K02's have brand new sensors)

My dealership quoted me $400/tire ($2K for all 5). I checked with a couple other places and they would install only their own brand. They also said they didn't use the Techstream software so they wouldn't install the Denso brand if I purchased them.
The light is flashing because one or more sensors aren't transmitting to the control unit. I have found out that if a tire with a working sensor goes down the light stops flashing and you know you have at least 1 low tire. So with my trusty air compressor I went to each tire, lowered the pressure to 20 psi and checked the light. If it stopped blinking I put it back to normal pressure and went to the next one. The L/R is the only one that didn't stop the blinking, so I know that one has the bad sensor.
Amazon has the sensors cheap enough but then there's the problem of getting the Techstream software and finding a shop to break down the tire, swap the sensor, air it up and check the balance. I need to check a few shops to see if they will do that, get the knock off Techstream from China off of Amazon and try to get that one working again. Then I'll do the others.

I just don't think TPMS is worth it in our trucks. If your light comes on all it tells you is at least ONE of your tires has lost pressure. Doesn't tell you which one, or what the pressure is.
In my case, the sensor in my spare was bad. Well my spare is seized up and have had no luck with being able to get it down. So even if I replaced all 4 sensors my light still would've been on.
My friend's 3rd gen Tacoma will show you the tire pressures of every tire so you can actually know which one is low. That's useful and worth having TPMS for.
If your TPMS light comes on while driving on the highway and you pull over there's no guarantee any of your tires will actually read low. It could just be your spare in which case you pulled over for nothing. Trust me, you'll know if you are quickly losing pressure from one of your 4 corners. You don't need a light for that.
Get some E rated tires and delete that light. Save the money and put it towards something more useful for your truck.

I agree that it could be a better system, but my T4R is 2004, bought in '03. Systems have become better and you can certainly upgrade to a newer TPMS system that will provide you with individual pressures, even tire temperatures.
I see it as more of a maintenance item than an emergency alert. I check my tire pressure regularly but I've had leaky tires before, so I like to be nagged if one needs to be aired up. I have also found it useful on trips, especially if I'm towing something. If the alert comes on I doubt that I would pull over unless I felt the handling was being impaired. I'd probably just wait for the next gas stop and do a walk around with my tire pressure gauge.
I really hate that Toyota service departments charge so much for this minimal information. I think $400 a tire is totally unreasonable for this service. I like the TPMS but not that much.
