Also make sure that you are stoppped and that it is in the correct position.

To engage the diff lock you must stop the vehicle,put it in N then push the 4wd lever to the right and then forward then put in drive and go on your merry way.
The is the "grey wire mod" which allows diff lock operation in any mode including 2wd.

That which I described is for the rear diff lock and if the Runner is equipped with one it will have that RR DIFF LOCK button to the side left side of the wheel.
Try getting into a gravel lot, put it in 4lo, engage the locker, and drive slowly in a serpentine pattern, like an S.
Try doing this in both forwards and reverse.
If the locker has been dormant for a while it will take a little nudge to engage
