I did a flush at 90,000 miles with my major service. Seems a lot of people don't recommend it, but the local Toyota Service recommended it. I just don't think that fluid with that kind of miles would do every thing we ask tranny fluid to do. It's been about 6weeks and 4,000 miles and no troubles so far. I know after the major service the truck runs better, and my shifts are smoother--even increased my gas mileage by 1mpg. Just my .02

Is there a major disadvantage/negative/objection/mistake of doing a simple drain and refill, then drive for a a few hundred miles and repeat the drain and refill 2-3 times so that you "dilute" the old fluid with new?
