I never drove a 4Runner with the V8, so I don't have any way of comparing the two......but my 2005 SR5 with the V6 has more than enough power for the driving I do. Whether I am merging onto a freeway such as I-95 and need to get up to 70 mph quickly or I need a burst of speed when a light turns yellow, the DOHC V6 does just fine.
I am a "gearhead" from back in the day and like V8 power as much as anyone, but did not find it necessary for my 4Runner. I also think it eats plenty of gas as it is -- and the V8 would be slightly thirstier. Besides, I like the idea of only using 4x4 when I need it; rather than having it on all the time as the V8 does.
My V6 does well on hills at high speeds, although it will "gear hunt" sometimes and shift in and out of overdrive when slightly straining against gravity. Now that can be annoying and usually results in me bumping the shifter down into "4" until I reach the top of the hill.
I-70 in Western MD and parts of the PA Turnpike are roads that always cause this with my 4Runner, but I only travel them once or twice a year. There is also a very high bridge that I cross on my way to work, and it can lead to "gear hunting" if you are going less than 75 mph on the incline. I suspect that a 4Runner with the V8 option would be able to hold the higher gear the whole way across. Oh, and the "V8" emblem on the grill does look pretty cool.

My synopsis is that if you are looking for a "street fighter" or want to drag-race every other car on the road, then don't buy a 4Runner at all. If you like the 4Runner for what it is, then get the standard V6 and enjoy it; as you will not be disappointed in the plentiful power. Should you be one who tows often, has five kids and climbs a lot of hills, then pony up the ducats for the V8 engine option.
Either way, you will not be sad. After all, it is a 4Runner!! I am quite satisfied with my V6, and you would be too. Then again, those who have the V8 say it was worth it - so well all win in my book.
