1. A locker is a differential locker. Normally, the rear differential is what we call "open". Meaning the wheel with the least resistance gets the power. So if one wheel is in the air, it will be the one getting power, and the wheel on the ground won't get any. Not good for offroading. WHen the differential is locked, both wheels get the same amout of power no matter what, even if one is in the air. E-locker's are the factory electronically controlled mechanisms that let you lock your diff with the push of a button. That way the diff is open for normal driving, but you can lock it when you are offroad. Air lockers are the main aftermarket lockers used. Search for ARB lockers on google to find out more about them.

2. Highlander was what the sporty trim model was called in 1999 and 2000. In 2001, they started calling them "Sport Edition". My truck is a Highlander, look at a couple pics and you'll see what it looks like. They also have the rear diff locker as part of the package.

3. First type is a suspension lift, in two flavors: a) spacers that go on top of your existing springs or b) new springs. The second flavor is a body lift, often abbreviated BL. Poly spacers are installed in the body mounts, to lift it off the frame. A body lift does not affect the suspension at all. Beyond that, you need to do a bunch of reading to find all the different brands and products you can use to do the aformentioned lifts.

Quicksilvr answered everything pretty darn good, but for your last question he seems to have forgot to mention the 3rd way of lifting your truck..
